The truck is said to have lost control and ran off the road into a ditch and onto the Ballarat-Maryborough railway line.
The driver could not be rescued and died on the spot. They have yet to be identified.
Police are investigating the accident and preparing a report for the coroner.
There are significant delays in the area and the western highway is expected to be closed in both directions for “some time”.
The highway was closed in both directions between the Midland Highway and Ballarat-Burrumbeet Road.
It reopened westbound just after midday, but lanes to Melbourne remain closed and detours remain in place.
The V/Line in Ballarat has also been disrupted and some services will come from Ballarat rather than Maryborough.
Anyone who witnessed the accident is asked to contact the police.
